• Mutagenized a potentially critical amino acid to the catalytic manganese cluster responsible for photosynthetic water oxidation of substrate water.• Purified the mutated Photosystem II variants using IMAC and SEC. Performed cellular and molecular bioassays to characterize the deleterious effect of the amino acid substitutions on enzymatic activity.• Trained fellow lab members in molecular biology, protein purification, and immunological methods. • Shared results, reagents, and procedures with external collaborators to further characterize the role of Arg347 in CP43 using EPR and other spectroscopic methods.
