Major responsibilities are: 1) Generated novel capsid libraries by insertion of wildtype AAVs to dose rodents and nonhuman primates2) Discovered and identified lead capsids through directed evolution of AAV libraries in primary brain microvascular endothelial cells, rodents, and nonhuman primates3) Performed various preclinical assays to characterize lead capsids of rAAV vectors by their transduction efficiency, tissue tropism, and biodistribution. 4) Produced, purified, and checked qualities of rAAV vectors in large scale. Major skills: rAAV vector design, production and purification, evolution of AAV capsid libraries in vitro and in vivo, biochemical characterizations of novel AAV capsid variants, real-time PCR, digital-droplet PCR, next-generation sequencing.
