ORBIT (Oncology Research for Biologics and Immunotherapy Translation), Institute of Applied Cancer Science, MDACC, Houston, TX• Successfully identified clinical antibody candidates from a panel of leads by cell-based immunoassays (ADCC, CDC, and MLR etc.) for a T cell immune checkpoint.• Leading a novel project targeting a myeloid cell target, designing antibody discovery strategy, and developing in vitro cell-based functional assays for biological validation and lead screening.• Established numerous in vitro cell-based functional assays (e.g. NF-AT/NF-kB/IRF reporter cells, MLR, Treg suppression, M1/M2 differentiation, Th cell skewing, CTL and NK cell killing assays, tumor/immune cell coculture etc.) to advance molecules from biological validation, hit-finding to lead optimization.• Established a Flow cytometry protocol (TIL and peripheral blood analysis) for animal model selection, in vivo pharmacodynamic biomarker analysis, and target expression in patients.

Departments of Pediatrics and Experimental Radiation Oncology• Worked with Merck, GSK, and Incytes for combining radiation with different immunotherapy targets for treating lung cancer in preclinical models. • Generated the first anti-PD1 resistant lung cancer line and syngeneic lung cancer model (published on Cancer research 2017).• Wrote several successful research proposals: (1) Combining anti-OX40 and radiation in preclinical lung models (funded by GSK, $280K); (2) Combining anti-PD1 and radiation in preclinical lung cancer models (funded by Merck, $200K); (3) Improving CAR T cell therapy for solid tumors (awarded by lung cancer research seed funding, $30K). • Teleconference meetings with outside collaborators (Merck and GSK). • Supervised two junior research assistants.

Dept. of Pathology & Immunology, Baylor College of Medicine, Houston, TX• Studied mechanisms of T cell receptor and B cell receptor signaling regulation and autoimmune diseases (PhD thesis: Regulation of lymphocyte signaling and function by kinase HPK1).• A deep understanding of T and B cell receptor signaling and myeloid cell biology.• Assisted with grant writing including several RO1 and R21 grants.

Dept. of Biochemistry and Molecular Biology, Tongji Medical College, Wuhan, China• Identified the upregulation of PD1/PDL1 mediated tumor evasion in a liver cancer mouse model. • Identified that chronic inflammation such as exposure to low dose interferon gamma induced PDL1 and promoted tumor growth.
