I work independently in the development of novel as well as implementation of established in-vitro Cell-based platforms for determination of immunogenicity of various therapeutic modalities (antibodies, nanobodies, peptides, antibody-drug conjugates, RNA-based drugs, gene therapy). I work independently as a part of research teams focusing on identifying and validating novel cancer drug targets and drug combinations. I utilize expertise in molecular and cellular biology, cancer biology, and functional genomics screens to execute and interpret experiments to develop targets for therapeutic intervention. Depending upon the stage of projects, my specific duties include:- Utilize molecular and cellular biology skills including DNA cloning, cell culture, lentiviral genome-wide shRNA and CRISPR screens followed by global next-generation sequencing analysis ; - Apply both genetic and pharmacologic approaches to modulate target candidates in vitro and assessing the functional impact on cancer cell biology. These include RNAi, CRISPR, small molecules, antibodies, and other pharmacologic approaches to modulate targets in vitro followed by molecular and phenotypic analysis;- Execute molecular and cell-based assays across a wide variety of technical platforms (qPCR, Western blotting, FACS, immunofluorescence, ELISA, etc.) to assess target modulation and its impact on cell viability and cell cycle disruption, downstream signal transduction, transcription and immune susceptibility; - Utilize expertise in cancer biology to design, execute, and analyze experiments to advance mechanism of action target biology and develop innovative approaches for targeting cancer; - Utilize electronic data capture/analysis tools such as Chalice, Excel, GraphPad, Biobook, Spotfire, and PowerPoint for experimental documentation and data analysis/presentations; - Present experimental results at internal group meetings.

I focused on profiling epigenetic inhibitors in mouse tumor cell models. I performed identification and validation of novel drug targets and drug combinations in the fields of cancer epigenetics and immuno-oncology.I worked independently and also as a part of team to design, execute, analyze, interpret, and refine experiments, including biochemical and molecular assays both in vitro and in vivo target modulation in model systems, in vitro pharmacology, biomarker assay development, and clinical sample analysis. My specific duties covered:- Develop, optimizing, and implementing assays that constitute the compound screening funnel. I generated engineered cell lines for primary and secondary pharmacology;- Implementing genetic and pharmacological screens to identify new targets and potential drug combinations;- Utilize 2D and 3D (spheroid models, soft agar, colon forming assay, 3D co-culture systems, etc.) cell growth assays to measure cell proliferation, migration, and invasion;- Execute molecular and cell-based assays across various technology platforms to assess target modulation and measure downstream signaling pathways– qPCR, IP, FACS, immunofluorescence, western blotting, ELISA, high content screening, etc;- Collaborate with in vivo pharmacologists to perform in vivo efficacy and biomarker studies;
