Conducted Ph.D. research under the supervision of Professor Frank Staal in the Stem Cells and Lymphocyte Development Group. Unfortunately, due to funding limitations, my PhD project could not be extended, which led to my inability to meet the graduation requirements at Leiden University. I didn't get the PhD degree. Nevertheless, I gained significant experience in CRISPR-Cas9 gene editing and T cell differentiation, focusing on two major projects:Developed a CRISPR-Cas9 loss of function model to study the role of T-cell-specific transcription factor 1 (TCF1) in human T cell development.Investigated the restoration of T cell development in Severe Combined Immunodeficiency (SCID) patients with RAG1 mutations using lentiviral gene therapy and artificial thymus organoids to monitor T cell differentiation, analyze T cell development blockages, and TCR gene rearrangement stages.Collaborated with multidisciplinary teams to enhance research outcomes and ensure project goals were met. Presented research findings at national and international conferences, raising the profile of the institution.
