Senior Group Leader

Current
Functional Genomics Consortium (Fgc), Broad Institute Of Mit And Harvard

- As part of the Functional Genomics Consortium (FGC, membered Abbvie, Merck, Johnson & Johnson), manages its production activities that include constructing various types of CRISPR libraries, CRISPRko/CRISPRi/CRISPRa, in pooled or arrayed format.- Leads the saturation mutagenesis efforts to (1) assemble screenable libraries (that is, in lentiviral expression vectors) that contain mutants of all possible single amino acid substitutions for a given ORF, (2) de-convolute screen samples from extracting ORF out of genomic DNA to preparing Next-Gen Sequencing (NGS) samples, (3) leverage expert-level R programming and Data Science skills (an “A” student <100% in fact> in Data Science when tried back-to-school to titrate against the best at Harvard in Fall 2018) to process/visualize the massive NGS data and produce hit lists.

Jan 2016 - Present

Manager Of Rnai Production And Development, The Rnai Consortium (Trc)

Cambridge, Ma, Us

- Pioneered the shRNA library production/development effort at the RNAi Consortium (TRC) to construct genome-scale lentivirus-based RNAi library targeting Human and Mouse. The TRC is a global project; its academic and corporate funding members include: Academia Sinica in Taiwan, Bristol-Myers Squibb, Eli Lilly, Johnson & Johnson, Novartis, the Ontario Institute of Cancer Research, and Sigma-Aldrich. - Led the efforts to build hORFeome V8.1, a high quality genome-scale human ORF Entry clone collection, and to create subsequently the first lenti-viral ORF Expression library that is amenable to mammalian gain of function screens. - Directed the efforts to build 'barcoded' (24 nucleotides) ORF Expression library that for the first time enables pooled ORFeome screens. Hands-on experience in arrayed and pooled screening. - Oversaw numerous R&D projects that enable the application of the shRNA library and hORFeome library in both loss- and gain-of-function genetic studies.- Manages 4 high throughput production groups including clone production team (shRNA, ORF, TALE, CRISPR etc.) , transfection-quality DNA prep team, lentivirus prep team and shRNA library knock-down validation team.- Established ORF mutagenesis pipeline as part of "Target Accelerator (TA)' efforts at Broad Institute.- Proficient in perl programing, SQL query, R programing, Python programming. Passionate for large data handling and analysis.- Take pride of being able to establish/run wet lab operations, optimize/automate/troubleshoot them, and conduct immediate data analysis, and enjoy it.

Jan 2005 - Jan 2016

Manager Of (Genome) Finishing Laboratory

Cambridge, Ma, Us

- Oversaw the expansion of the Genome Finishing lab from 5- to 25-member team throughout the human/mouse genome sequence finishing projects.- Established high throughput workflows vital to the completion of Human and Mouse genome sequences: transposition bombing, various versions of PCR methods, shatter library, special sequencing chemistries. - Led automation efforts, both robotically and informatically, to serve the high throughput activities.- Automated tracking and reporting systems by database queries and scripted software for lab daily applications.- Instilled the team a great work ethic that proved to be essential in answering the challenges brought by high-profile projects.- Successfully conducted pilot project for the Institute’s efforts to build gemone-scale shRNA library targeting both Human and Mouse. This pilot work paved the way to eventually establishing the RNAi Consortium (TRC).
