Research Experience:Project Title: Regulation of Prostate Epithelial Homeostasis(1) Investigated the role of CD133 (Prominin-1) as a putative prostate epithelial stem cell marker and its function in the prostate development.• Experimental design, troubleshooting, experimental notes writing, and manuscript writing under the mentorship.• Execute the experiments, including transgenic mouse models, mouse survival surgery, RNA in-situ hybridization, prostate sphere/organoid culture, gene cloning/subcloning, flow cytometry and cell sorting, qRT-PCR, cytospin and immunofluorescence staining, immunohistochemistry, western blots, and data analysis. (2) Studied the role of stromal Wnt signaling in prostate stem cell niche.• Experimental design, troubleshooting, experimental notes writing, and manuscript writing under the mentorship.• Execute the experiments as the team lead with two postdocs, including transgenic mouse models, human prostate FFPE specimen preparation, in vitro and in vivo small molecule analysis (BrdU, tamoxifen, IWP-2, Wnt3a, Wnt5a, Tgfb1/2/3), primary/prostate cancer cell culture, flow cytometry and cell sorting, RNA/DNA purification and qRT-PCR, RT2 Profiler PCR array, laser-capture microscopy, gene overexpression/knockdown, prostate sphere/organoid culture, mouse survival surgery, RNA-sequencing, CHIP-PCR assay, western blots, histology and immunostaining, confocal microscopy, and data analysis.
