ELISA, Western Blot, Immunization, Protein Purification, and Molecular Biology

1 Performed biochemical analysis of an extremely low-expression membrane protein. Optimized different conditions in Co-IP and western blot to successfully purify the protein complexes and identify its binding proteins through LC-MS/MS.2 Performed RT-PCR to get cDNA from animal tissue. Constructed expression plasmid to express proteins in mammalian and insect cell lines; tested the protein function with cellular assays such as calcium imaging, and lastly lysed cells and detected proteins with western blot.3 Performed ELISA to screen for drugs that disrupt an antigen-antibody interaction.4 Used standard molecular biology techniques, including Gibson assembly, primer design, and site-directed mutagenesis to design and construct plasmids to generated CRISPR-cas9-guided knock-in flies which are a reporter line of a protein.
