• Serve as the clinical biomarker lead for multiple T-cell engager (Alnuctamab) and CAR T (ide-cel and liso-cel) programs ranging from Phase I exploratory trials to Phase III registrational trials.• Partner with discovery groups to develop biomarker strategies in preparation for IND application and FIH studies of assets in solid tumor.• Design and direct biomarker plans, work with CROs to ensure execution of biomarker assays, generate and deliver biomarker data to support clinical decision making and pre-clinical pipeline development.• Collaborate with various functions (clinical operations, pharmacology, biostatistics, etc.) to author clinical documents such as protocol, statistical analysis plan, response to information request from health authorities and clinical study reports.• Lead discovery projects with bioinformatic groups to analyze and interpret clinical biomarker data. Communicate translational learnings with internal pre-clinical teams and external collaborators to advance understanding of diseases and to guide development of next generation immuno-oncology assets.

• Led discovery projects to unravel BiTE mechanisms of action, novel biomarkers and combination strategies.• Designed and conducted genome-wide CRISPR loss-of-function and CRISPRa gain-of-function screens to identify resistance mechanisms of cancer cells to BiTE-mediated T-cell cytotoxicity. Independently performed data analysis, target identification and validation.• Conducted in vitro T-cell cytotoxicity assays and in vivo efficacy studies to demonstrate perturbation of multiple co-stimulatory ligands and apoptosis pathway as cancer cell intrinsic mechanisms to evade T cell engager therapies.

• Directed an independent project to investigate the function of KLF4 and its downstream MAP2K7-JNK signaling in T-cell acute lymphoblastic leukemia.• Initiated the generation of PDX models in the lab and utilized the models to evaluate effects of small molecule inhibitors targeting JNK pathway on human leukemia progression in vivo.• Established and expanded the Patient-Derived Xenograft (PDX) repository in the lab to further study essential signaling pathways in T-cell leukemia in vivo. • Cooperated with lab colleagues to study the development of T cells in thymus, and homeostasis and differentiation of CD8+ T cells using loss-of-function mouse models.• Spearheaded multiple collaborative projects with TCH clinical teams, industry, and international institutes for pre-clinical testing of novel therapeutic approaches for leukemia patients.• Self-trained on GeneSpring and analysis of high content datasets (GEO, ArrayExpress, Oncomine, etc.).• Developed and optimized multiple standard operating procedures (SOPs) in the lab.• Supervised the training of junior colleagues and recruited two graduate students to the lab.

• Utilized a novel knockout mouse model in combination with flow cytometry, in vitro functional assay and immunoblot to study the role of a phosphatase PP6 in T cell development and function. • Independently performed frozen sections and immunohistochemistry staining to investigate the expression pattern of a vacuole protein VPS18 in mouse brain.• Extensively collaborated with lab colleagues and co-authored two peer-reviewed papers.
