In vitro Compound Screening of BPTF Protein Binding Ability with Mono-phosphorylated H3K4 Peptide.- Bioinformaticists in the Drug Docking Group designed possible compound in silico that could assist target protein to bind with abnormal phosphorylated peptide in cancer cell- E.coli BL21 Rosetta 2 cell culturing with antibiotics screening- IPTG induction for expression of customized pGEX6p-1 GST-BPTF vector to produce GST fused BPTF protein- Used multiple methods (sonication, dry ice-ethanol freeze and thaw) to lyse cells and purified GST-BPTF protein through Glutathione beads column. - Concentrated and dialyzed purified GST tagged BPTF protein; carried out compound pull-down assay of GST-BPTF protein with phosphorylated peptide by using Dynabeads; and then used PreScission Protease to cleave GST tag off from unused protein sample- Superdex 75 gel filtration columns (FPLC) was then used to collect pure BPTF protein for protein structure determination- Collaborated with NMR and Mass-Spec facilities at UBC and SFU to discover compound structure, protein structure, and compound-peptide- protein binding.
