My research interests lie in protein-protein interaction mechanisms. I am especially interested in protein-protein interactions playing regulatory roles in cell signaling pathways which are mostly related to development, ocular, and degenerative auto-immune diseases. The major challenges in understanding the interaction mechanism are their promiscuous but context-dependent nature and the complex networking of signaling pathways. I am trying to elucidate the challenges by utilizing various structural biology techniques (Crystallography, NMR, SAXS, and Cryo-EM), thermodynamic assays, and computational tools (Bioinformatics, Molecular dynamics). Once understanding the mechanisms, one can design targeted drugs derived from peptides and implement a therapeutic system to patients through systems biological approach.

This is the very first study showing positive regulationon PDZ - Syndecan interaction upon phosphorylation.● Successfully designed a PDZ domain mutant with desired peptide ligand specificity.● Confirmed newly identified cell polarity complex interaction (Scribble - SGEF) in vitro anddetermined a novel PDZ – internal PDZ binding motif interaction mechanism.● Determined 16 noble protein crystal structures (6 Apo structures and 10 ligand-bound complexstructures). Assigned 2 noble protein NMR HSQC profiles.● Developed structure based sequence alignment and structural motif extraction tools (computersoftware written in JAVA) for PDZ domains.● Initiated collaborative effort to utilize molecular dynamic simulation to design artificial PDZdomains de novo with Dr. Thomas Simonson, Ecole Polytechnique, France.● Collected experimental data and performed experimental validations to utilize polarizable forcefield molecular dynamic simulation to predict and design PDZ binding peptides, in collaboration with Dr. Michael Schnieders, University of Iowa, USA. ● Established membrane lipid bilayer mimicking Nanodisc system in Fuentes Lab. -
