Study DNA secondary structures such as DNA hairpins, and DNA modifications such as 5-methyl-C by protein nanopore (third generation DNA sequencing technique), HPLC, Uv/vis, CD, and MS. Internal hairpins and fishhook hairpins were found to have different unzipping mechanisms and locations. Explore the latch zone of alpha-hemolysin with a group fishhook hairpin structures. Besides abasic site, current differences between native base pairs were detected.Investigate the interactions of DNA hairpin with groove binders such as polyamines by protein nanopore. The binding of Spm and Spd were so strong that the hairpins will not unzip, while Put does not have an effect on the unzipping of hairpins.Demonstrated the kinetics of different length of CAG repeats are different even though they have similar thermodynamic stability.
