My undergraduate research project, titled "Optimizing CRISPR Delivery and Guide RNA Engineering for GalNAc-Targeted Gene Editing." Validating gene editing efficiency and specificity of GalNAc gRNA-mediated CRISPR delivery in liver and reporter cell lines. Evaluating and optimizing various CRISPR transfection methods using Cas9/gRNA plasmids to enhance gene editing efficiency. Engineering optimized GalNAc-targeted gRNAs and experimental conditions to achieve breakthrough results in gene editing efficiency and precision by flow cytometry and fluorescence microscopyMy undergraduate research project, titled "Guide RNA Engineering for Targeted CRISPR Ribonucleoprotein Delivery," was presented at the SB3C2024 conference. All the research was done in Dr. Yeh-Hsing(Sima) Lao's lab. Wet Lab Technique: 1) Proficient in mammalian cell and bacterial culture, cloning, plasmid and DNA/RNA extraction, transfection, immunostaining, lentiviral preparation and transduction, FACS, PCR, and qPCR.2) Execute operating flow cytometers, fluorescence microscopes, and confocal microscopes, automated cell counters.3) Applied design of experiments (DOE) techniques to optimize experiments and improve process outcomes.4) Specialize in analyzing data using Microsoft Office, R Studio, Prism, Fiji (Image J), Flow Jo, and Qu Path.
