Accomplishments: Developed an automated, gentler passaging method on a Hamilton liquid handling robot for shear-sensitive cell lines. Applicable for high throughput media development for speed and data consistency of adherent and suspension cultures.Developed a modified coating reagent for the coating of cultureware to limit the adherence of "sticky" cell lines for non-adherent cell culture work.Co-developed an easy-to-use staining device for immunocytochemistry staining of 3D aggregates/organoids with preserved yield and homogeneous staining.

The integral part of my thesis in the Haynes lab (under the supervision of Dr. Charles A. Haynes) is to perform the selection of aptamers (i.e. short single-stranded DNA) against multiple targets using a platform known as High-Fidelity SELEX. I have successfully selected high affinity pool of aptamers against thrombin, mesothelin, and andexanet alfa. Throughout my laboratory experience, I've gained several biotechnology skills such as performing PCR, real-time PCR, digital PCR, gel electrophoresis, DNA extraction, diversity determination assay, and affinity determination assay. The end goal of Hi-Fi SELEX is to use these aptamers on applications such as decorating nanoparticles with aptamers for efficient drug delivery and immobilization of aptamers as ligands for purification of protein and small molecules.

Project title: Definition of yeast strains in order to find superior starter cultures for beer brewing.My responsibilities include preparing agar and growth media for the growth of 12 different yeast strains and Lactobacilli that may be present in the yeast media. I'm capable of operating an autoclave and using aseptic technique for inoculation and isolation of yeast. The DNA of the isolated baker's yeasts are extracted using glass beads and boiling. The extracted DNA are then amplified using "Random Amplified Polymorphic DNA" (RAPD) using 3 different primers, which are P73, O-PA1 and O-PA10. After performing gel electrophoresis, the optimized DNA extraction method and type of primer used are determined. They are then employed on the isolated of the 12 yeasts strains. The genomically defined strains are evaluated for efficiency and quality ensuring characteristics in a micro brewery.
