Postdoctoral Researcher
The aim of my project is to understand the roles and the regulation of tetraspanin 8 (tspan8) in prostate cancer (PCa). This transmembrane protein is involved in cell adhesion and migration and is linked to cancer stem cell activation. The first objective of our research is to validate tspan8 expression as a biomarker of cancer aggressiveness and dissemination in a cohort of PCa patients. Then, we want to understand how tspan8 expression is regulated in PCa. We are developing a siRNA screening approach to identify the regulators of its expression in a cellular model of PCa. Finally, we are testing antibody fragments (single-chain variable fragment) conjugated to a strong antimitotic drug to target specifically tspan8-positive tumors.

At HalioDx, my role was to contribute to the development of qPCR kits in the field of oncology for an external client. I worked on the clinical documentation of these kits to comply with the European In Vitro Diagnostic Regulation (IVDR) and performed literature monitoring in relation to the biomarkers of interest.

Research Intern In Molecular Biology
Plasmid DNA production for transient protein expression in mammalian cellsWithin the Novartis Institutes for Biomedical Research, the Protein Production and Antibodies group is responsible for providing antibodies and antigens for research purposes. In order to produce both antibodies and antigens in mammalian cells, plasmid DNA (pDNA) carrying the gene encoding for a protein of interest are used. Large amounts (2-50 mg) of high quality and purity pDNA are needed for each mammalian transient transfection and protein production. The current method for pDNA production has a relatively low yield compared to the requested amounts and is also expensive. A new approach for pDNA purification was developed and the purposes of the project were (1) to define the best transformation and bacterial culture settings for pDNA production, then (2) to test, improve and characterize the new method for pDNA extraction and purification and finally (3) to control the produced pDNA for transient protein expression in mammalian systems.

Sensitivity to AICAR and thiamine metabolism in Saccharomyces cerevisiaeAICAR is a monophosphate intermediary metabolite of the de novo purines synthetic pathway presenting very interesting potential antidiabetic and antiproliferative properties. It has also been show that AICAR injection to mice induces an increase of muscular mass in the absence of exercise. Finally, accumulation of this metabolite in cells is responsible for severe genetic diseases. However, the molecular mechanisms involved in physiological, pathological and therapeutic effects of AICAR are still poorly understood. The laboratory has identified and characterized some of these molecular mechanisms in a unicellular eucaryotic model, the yeast Saccharomyces cerevisiae. A genetic screen previously performed allowed them to isolate around 30 yeast mutants hypersensitive to AICAR accumulation. Among those, three of them presented a missense mutation in a thiamine metabolism encoded gene, as revealed by next generation sequencing of their cognate genomic DNAs. The goal of this placement was (1) to validate the genetic link between the identified mutations and the AICAR hypersensitivity and (2) to understand by several approaches how these mutations lead to AICAR hypersensitivity. The results allowed us to demonstrate that thiamine carriers are responsible for AICAR riboside uptake in yeast.
