Microbes are intimately involved in our health and the health of marine ecosystems; yet, we are far from the level of understanding needed to implement efficacious intervention strategies using microbes in either the human body or marine ecosystems. I was specifically involved in studying the mechanisms by which the bacteria Pseudoalteromonas luteoviolacea instigates metamorphosis, life-cycle transition from a motile larva to a sessile polyp, of the tubeworm, Hydroides elegans. Factors, comprised of phage-like protein syringes, produced by the bacteria bind to, and activate, metamorphosis in the tubeworm. All lab research centers on this paradigm.My first collaborative project involved designing, testing and verifying expression vectors containing different constitutive and experimental gene promoters for use in Pseudoalteromonas luteoviolacea. My most important contribution was developing the luciferase assay to analyze the strength of promoter expression using a luciferase (light) reporter. My second ongoing project was investigating a putative transcription factor in Pseudoalteromonas luteoviolacea. This work involved characterizing deletions of the gene encoding for the transcription factor, dubbed Metamorphosis-associated contractile regulator (MacR). Deletions were demonstrated to reduce the rate of metamorphosis in the tubeworm and the level of expression of gene promoters upstream of metamorphosis genes in Pseudoalteromonas luteoviolacea. Current work involves characterizing the entire collection of targets affected by MacR using RNA-Sequencing methods. However, due to personal stress arising from the war in Ukraine and burnout from almost six years of graduate school, two of which were spent during the latest pandemic, I have decided to take a leave of absence for at least the subsequent term. I do not plan to forsake graduate studies and expect to resume studies upon resolving the personal issues that stymied progress.
