About Ami Yamamoto

Expert in ex vivo and in vivo modeling of cancer and metastasis, tumor organoids, and cell biology. I am passionate about effective communication with both scientists and non-scientists. In my PhD work, I developed a rat model of breast cancer metastasis to efficiently study circulating tumor cells (CTCs). I revealed that there is a temporal association with CTC abundance, formation of a necrotic core in the tumor, and the increase in dilated blood vessels. I further identified Angptl7 as a tumor-derived, necrotic core-enriched secreted protein and found that suppressing Angptl7 reduces CTCs, necrosis, metastasis, and dilated blood vessels. The work identified Angptl7 as a therapeutic target for breast cancer metastasis and also shed light on "the importance of what is on the inside" of tumors, revealing the contribution of the tumor necrotic core in metastatic dissemination.https://www.pnas.org/doi/10.1073/pnas.2214888120
