Skills:• Extensive hands-on experience (>8 years) (independent operation, maintenance, and optimization) in analytical method development/ validation for chromatography (basic reverse phase, strong cation exchange) and MS. Analytical instrumentation including LCs (nanoLC, UPLC, HPLC) and Mass Spectrometer (Sciex-ESI MS/MS-Triple-TOF 5600, Sciex- QTrap 6500, Thermo Scientific-Orbitrap Velos Pro, Q Exactive HF, Orbitrap Eclipse Tribrid MS.• Extensive proteomics (Body fluids, like Plasma/ Serum, Saliva, HVS, Hair, Tissues, Cells, etc.)- including proteome profiling, protein-protein interactions, and PTMs (Carbonyl, Phospho, Glyco, etc) using IDA/DDA; HCD/ ETD, label-free (DIA (SWATH), Spectral Count, Peak Area); and labelled (iTRAQ-4plex and 8plex, TMT) quantitative; targeted acquisition (MRM); AP/ IP-MS; Peptide Mapping; and De-Novo sequencing. • Skilled in designing and implementing experiments within strategic projects. sample preparation, generating reproducible quality control data, research article writing and documentation, and teamwork.• Comprehensive data analysis: MS-based data analysis and statistics workflow using different software like Analyst, Protein-Pilot, Mascot, Peak View, Marker View, Multiquant, Xcalibur, Protein Discoverer, Maxquant, Spectronaut pulsar, Skyline, PEAKs, Peptide-Shaker, Cytoscape, Network Analyst, Webgestalt, DAVID, Panther, STRING, Reactome and KEGG pathway.     I have been involved in the MS ‘OMICS’ (Orbitrap Velos Pro) approach to decipher the functional biology in malaria (Plasmodium falciparum) and tuberculosis. Recently I engaged to characterize PTMs, especially N-glycosylation, Methylation, Acetylation, and Phosphorylation of the Plasmodium surface proteins; and to profile food vacuole proteins in the role of malarial drug resistance of the erythrocytic life stages of the parasite. I have also engaged to decipher multi-drug resistance (MDR) of Mycobacterium from clinical isolates by a quantitative proteomics approach (TMT).

    • I engaged in clinical proteomics for biomarker discovery and validation (Triple-TOF 5600+, SCIEX; QTRAP-6500+), and deciphering molecular events in pregnancy. For early prediction of preterm birth, we profiled (SWATH-DIA-MS and MRM methodology) an expressional trajectory for modulated proteins in maternal Plasma/ Serum, Saliva, HVF (high vaginal fluid), etc. in term/ preterm pregnancy within garbhini cohort (https://www.garbhinicohort.in). We first time demonstrated longitudinal maternal saliva/ HVF proteome changes in normal pregnancy. The discovery proteomics (in nested case (preterm)-control (normal) design) of the Plasma is over, now the validation part is going on.• Interestingly we also investigated the correlation between stresses measured by hair cortisol level on plasma protein ‘carbonylation’ (Biotinylation, Streptavidin Enrichment, DDA) in pregnancy (in nested case (preterm)-control (normal) design). This is the first study to evaluate the carbonylation pattern of the proteins in plasma caused by perceived stress during pregnancy measured by hair cortisol levels. • I also involved in other collaborative projects like the role of wild-type p53 amyloids leading to cellular transformation and tumor formation in mice, global proteome changes/ DUBs profiling (MRM) due to lipotoxicity in hepatocytes (NAFLD model), the role of angiomotin in global protein synthesis in trophoblast cells, redox homeostasis under dehydration in rice, etc.

    Elucidated host-pathogen interactions in host cell adhered- Vibrio cholerae on virulence and regulatory gene expression (QPCR) by developing ‘adherence assay’ using human cell lines (INT 407, Hep-2, HeLa, undifferentiated and differentiated HT-29). This is the first study to demonstrate that ‘host cell contact’ is a signal regulating intracellular second messenger (c-di-GMP) levels via a three-component signal transduction system (VieSAB) and subsequent virulence gene (cholera toxin) expression in bacteria.
