During my internship at Laboratory of Molecular Physiology and Neurobiology I have experienced the following:- sterile work with hood, keeping murine embryonic stem cells undifferentiated anddifferentiating them to cardiac pacemaker-like cells;- handling 2-3 days rats and isolating neonatal ventricular myocytes to prepareprimary cultures employed to transfect plasmid DNA;- engineering of murine embryonic stem cells through electroporation;- extraction of DNA and RNA from singular cells and tissues and quantification ofnucleotides through spectrophotometer;- employment of DNase and retrotranscriptase, amplification of DNA/cDNAsequences through PCR and electrophoresis;- cloning of plasmid vectors, transformation of plasmid DNA in competent cells andpreparation of bacterial cultures.- Western Blot, immunofluorescence and flow cytofluorometry
