Thesis: "Campylobacter jejuni colonisation of the chicken gastrointestinal tract - mechanisms and prevention"Campylobacter jejuni is the leading cause of food-borne bacterial diarrhoeal disease in developed countries. This bacterium is common in the gastrointestinal (GI) tract of poultry, and poultry meat is regarded as one of the main sources for infection in humans. In order to develop new intervention strategies, we need knowledge about the C. jejuni colonisation in chicken intestines. In this thesis, we have therefore developed new DNA-based strategies for investigation of both the pathogen C. jejuni and the microbiota. These methods were used to study the colonisation of C. jejuni both in natural infected broiler flocks and in a mixed strain infection in broiler chickens. One of our main findings was that the microbiota of chicken GI tract did not affect the colonisation of C. jejuni. We identified a shift of the dominant C. jejuni strain over time, and this shift was the same despite different microbiota. These results lead to the hypothesis that it is the chicken immune response and not the microbiota that influence the colonisation pattern of C. jejuni in the chicken GI tract. Another finding was a C. jejuni colonisation of the crop mucosa, which reveals the crop mucosa as a potential reservoir for establishment of a C. jejuni colonisation. We have also observed a correlation in colonisation between C. jejuni and the chicken pathogen Clostridium perfringens. Our conclusion with respect to this correlation is that Cl. perfringens infection would make the chickens more exposed for a C. jejuni infection. For future development of intervention strategies, our results clearly point towards the upper GI tract. In this thesis we have preliminary data indicating that prevention of crop colonisation also prevents the colonisation of the lower GI tract.
