Research summary: Grapevine leafroll disease (GLD) is a major virus disease affecting Washington’s grape and wine production. To understand the etiology of the disease: I have sequenced the complete genome of two virus isolates, studied the 5’ non coding region, and identified the viral silencing suppressors. Epidemiology of GLD, including the spread of specific variant groups of the virus was monitored in the vineyard. Additionally, I have standardized a new technique to clone large genomes of plant viruses such as ampeloviruses and tospoviruses in fewer PCR steps. I also designed new primers and standardized protocols to detect two major grapevine viruses in a single multiplex RT-PCR assay.

Transgenic tobacco plants were developed that constitutively express PR genes responsible for defense against the nematode. High expression lines of transgenic tobacco produced less number of root knots and were resistant to nematode infection compared to the wild type control plants.
