Investigated the dynamics of endothelial calcium (transient activity and intercellular Ca2+ waves) and its relationship to vasodilation in blood perfused arterioles.Utilizing intravital video-rate confocal microscopy, together with cannulated and intraluminally perfusion to study microvessels. Developing custom IDL routines for the analysis of calcium dynamics and handling large (20GB) video data sets.Collaborated with Cornell University in the development of routines for semi-automated measurement of vessel diameter using a Web-base system for analysis and mass storage.

Investigated the role of mitochondria, iron and intracellular pH during human cytomegalovirus (HCMV) infection. Received an American Hearth Scientist Development Grant to pursue these studies.Demonstrated that the host cell increases iron and acid extrusion in response to viral infection. Collaborated with Cornell University in the development of routines for automated quantification of 3D mitochondrial morphology (e.g. number and length). Setup microscope fluorescence imaging system for multi-wavelength time course studies of living cells, and two-dimensional gel electrophoresis system to investigate the effect of HCMV on mitochondria proteins.

Investigated the effect of human cytomegalovirus (HCMV) infection on host cell ion transporters. Developed a custom microscope imaging system (hardware, software, specialized chambers) to study virally infected cells.

Investigated the effects of antipsychotic agents on calcium and cell volume regulation in cultured nerve cells.Set up a Universal Imaging MetaMorph/MetaFluor Imaging System and my custom, low photodynamic damage, microscope photometer system.

Investigated the effects of osmotic perturbations on sodium and chloride transport across cultured kidney cells. Demonstrated important roles of cell volume and calcium in regulating ion transport in these cells. Designed and constructed a specialized micro-Ussing chamber, and a fluorescence system for multi-parameter studies that has been utilized in five other laboratories.
