Purification Development Co-Op
Utilized a Quality by Design approach (QbD) to map out design space for single-use Cation Exchange (CEX) membranes. Performed a Design of Experiment (DoE) for bench-scale CEX membrane separations. Collected data and modeled profiles for Host Cell Protein (HCP), Higher-Order Aggregate (HOA), dimer, and yield to determine acceptable operating condition ranges. Evaluated regeneration capability of CEX membranes to assess impurity clearance performance over a cycle lifetime using the operating conditions determined from the DoE. Presented development work for this project to the purification group and suggested future applications of single-use CEX membranes within downstream processing.Exposed to pilot-scale downstream processing, assisting with several pilot-scale harvests and supporting Anion Exchange Chromatography (AEX) and Hydrophobic Interaction Chromatography (HIC) operations. Supported bench-scale polishing chromatography (AEX and HIC) development work with AKTA Avant liquid handling systems and UNICORN software. Assisted with size variant isolation of HMW and LMW species from Drug Substance (DS) using Size Exclusion Chromatography (SEC) to support protein characterization studies that are to be submitted as a part of Biological Licensing Agreements (BLA's). Independently operated bench-scale Tangential Flow Filtration (TFF) systems for enrichment of HMW and LMW species.Exposed to pilot-scale column packing and qualification and independently packed and qualified bench-scale columns with multiple resins. Operated independent pH and conductivity load adjustments to be used for late-stage development work. Additional responsibilities included daily pH and conductivity probe calibration, weekly buffer preparation, and distribution of lab consumables. All development work was recorded on an Electric Lab Notebook (ELN) and consumable use was tracked through a Laboratory Inventory Management System (LIMS).
Research Intern
Developed new nitrogen-doping protocols for treating Carbon Nanotubes (CNT) to increase conductivity while maintaining tensile strength. New methods lead to an increased areal loading of active material by 150% and doubled the specific energy storage of the devices. Co-authored an article detailing the findings of the new doping method which was accepted by Carbon journal in 2019, titled "Nitrogen-doped CNT on CNT hybrid fiber as a current collector for high-performance Li-ion capacitors."Utilized Scanning Electron Microscopy (SEM) to observe affects of autoclave time and temperature on CNT fibers and Raman spectroscopy to analyze changes in chemical composition of the fiber. Exposed to methods of Chemical Vapor Deposition (CVD) and its applications within carbon nanotube devices.
