Postdoctoral Associate In Infection Biology At The Biozentrum
I've developed a correlative light and EM (CLEM) technique at C-CINA, Basel, Switzerland and employ a stage onto which I can load ultrathin samples attached to EM grids to visualize them directly in the fluorescent microscope. I can then map areas of interest, say on GFP labeled proteins and then load the same grids into the transmission electron microscope for high resolution imaging and/or electron tomography. This allows to see spatial distributions and the inter-relationships between various cellular components.I am currently applying this technique in a variety of biological problems.Additionally, I am studying Salmonella that cause diarrhea or a more life-threatening systemic disease called typhoid fever. During typhoid fever, spleen and liver are appointed to act as major target organs, in which Salmonella reduplicate mostly intra-cellularly in phagosomes of infected macrophages. Cell culture infection studies propose that some Salmonella escape from the phagosomes to the host cell cytosol where they can be killed in a process involving host cell degradation (autophagy/xenophagy). However, the frequency of such events in vivo still remains unclear.Therefore, I am using the unique capabilities of the serial block-face scanning electron microscopy (SBF-SEM) technique to investigate host / Salmonella interactions simultaneously at the tissue, cellular level in a mouse typhoid fever model. Such large 3 dimensional data sets will provide a unique basis for unprecedented comprehensive analysis of host-pathogen in vivo interactions.

In my Ph.D. project, I studied the entry pathway of the Vaccinia virus with high-resolution cryo-electron microscopy. We described a new macropinocytic mechanism of extracellular enveloped virus and I worked on the Vaccinia virus early core activation and uncoating.
