Using tissue culture, molecular biology, and bioinformatics, we are examining whether LINE-1 retrotransposons contribute to recombination-based mechanisms of telomere maintenance. Using a plasmid based rescue system we are recovering LINE-1 integration events from tumors lacking telomerase. By analyzing the sequences of both pre- and post-integration sites we can determine the integration structures of the elements, which will determine whether L1 integrates into telomere repeats in tumors without telomerase.

To build upon our project from 2012, using the fim system from E. coli, which flips a promoter containing segment of DNA, this project sought to utilize synthetic biology techniques to engineer the fim switch by replacing the native promoter with another promoter and creating a uni-directional switch. We demonstrate that it can function as a reliable and efficient biological transistor, or “transcriptor”. Beyond storing information and performing basic computations, the system would serve as a very useful, tightly controlled switch. I helped present our project at the iGEM regional competition and our project advanced to the championship competition.

Using molecular biology and bioinformatics we sought to characterize a member of the Polyamine Uptake Transporter family of the plant pathogen Phytophthera Parasitica. The gene was codon optimized for expression in yeast and then a combination of toxicity and radioisotope uptake assays were used to assess the transport activity of this gene. Results were presented in a poster session at the 2013 Oomycete Molecular Genetics conference in Monterey, California.

By using the unidirectional recombinase HbiF to augment the existing recombinase system that relies on the unidirectional recombinase FimE, we designed a heritable, binary memory system that can be used as a bi-directional component in more complex systems. I helped present our project at the iGEM regional and championship competitions.
