Through the Northwestern CounterACT Summer Research Program, I will delve into research work, clinical observation, and informational sessions on toxicology, chemistry, immunology, nanotechnology, ophthalmology, and dermatology for an eight-week, full-time, paid program, continuing my research with TIGIT and FoxP3 as potential ways to stabilize regulatory T cells.PI: Dr. Caroline Le Poole

In this project, we introduced FoxP3 and TIGIT genes into DNA constructs to see if these Treg-promoting genes have lasting effects in stabilizing Treg function. Initially, the construct we developed contained both the TIGIT and FoxP3 genes, which proved problematic as the construct was too large for effective introduction into T cells. We decided to create two constructs that only had one of the stabilizing genes to compare and contrast their efficacy to maintain a regulatory phenotype of host T cells.PI: Dr. Caroline Le Poole

Using gene editing techniques based on CRISPR, we studied the impact of TET enzyme removal from hematopoietic stem cells in mice. Our study built on the findings of Dr. Vipul Shukla, whose research has shown that TET deficiency disrupts mature B cell homeostasis, leading to oncogenesis characterized by the accumulation of G-quadruplex and R-loop structures.PI: Dr. Vipul Shukla

In this project, we aimed to sequence bacteria under the chin region using Polymerase Chain Reaction (PCR) and gel electrophoresis. Antibiotic susceptibility testing revealed different resistance patterns among the identified bacteria, highlighting the importance of understanding antibiotic resistance in microbial communities.
