Project 1: Identifying biomarkers during breast cancer metastasis• Efficiently mapped the epitope region of SUMO-1 and SUMO-2/3 against their respective monoclonal antibodies using several rounds of molecular cloning, peptide purification and Immunoblot.• Designed and developed a novel technique called SUMO ePItope Peptide Elution (SUMO-e-PIPE) involving SILAC labeling, immunoprecipitation, Western blot and LC-MS to identify biomarkers in breast cancer metastasis.• Evaluated huge data sets from mass spectrometry analysis and created protein interaction maps using STRING software.• Resolved a network of genes and found to be involved in cell migration, stress response and glycolysis, implying that dysregulation of SUMOylation leads to cancer metastasis and tumorigenesis.• Executed 3D collagen cell migration assay to enforce that SUMO-2/3 is essential for migration and metastasis.• Authored and published two peer reviewed research articles and several poster presentations in international conferences.Project 2: Essential roles of SUMO in nuclear transport and cell division • Spotted and documented annulate lamellae pore complex primary neurons and smooth muscle cells using immunofluorescence microscopy.• Designed and engineered artificial Nuf2-SUMO-1/2 fusion constructs of different lengths by sub-cloning DNA fragments and site-directed mutagenesis.• Discovered that polymeric SUMO-2/3 chain modification of Nuf2 is required for CENP-E localization to kinetochore and chromosome alignment to metaphase plates by overexpression, knock down and immunofluorescence assays.• Employed biochemical techniques like in vitro binding assay and co-immunoprecipitation assay to claim that Nuf2 binds to CENP-E with more affinity when it is fused with SUMO.
