Taken together the previous… Show more In brief:- F. nucleatum, an anaerobic opportunistic pathogen has been well known for its role in oral and gut infections such as periodontitis and inflammatory bowel disease (IBD). Its association with CRC progression was just then identified by different research groups but, the mechanism underlying this progression was not revealed. Hence, I predicted the possible interactions between host and F. nucleatum followed by enrichment analysis of host proteins. Taken together the previous experimental studies, I hypothesized that F. nucleatum infection might enhance CRC progression by regulation of signaling cascades such as wnt/β-catenin pathway which lead to up-regulation of proinflammatory responses, oncogenes, modulation of host immune defense mechanism along with suppression of DNA repair system. With insights into the key role of F. nucleatum in several diseases, I predicted a set of druggable target proteins by implementing subtractive genomics studies. Among them, MurA enzyme that catalyzes the first committed step of peptidoglycan synthesis pathway was selected for in silico and in vitro inhibition studies. Orientin and quercetin-3-O-glucuronide (Q3G) were identified as potential inhibitors based on structure-based virtual screening (SBVS) and molecular dynamic (MD) simulation analysis. Simultaneously, I cloned the murA gene from F. nucleatum into BL21 cells and purified MurA protein with affinity column chromatography. These two compounds inhibited MurA activity in vitro with inhibitory constant (Ki) of 51.09 ± 2.06 and 9.53 ± 1.58µM, respectively. Simultaneous studies by other research group showed the inhibitory potential of Q3G on bacterial infection, which suggest that the compound can well pass through various membrane barrier and has antibacterial activity as well.
