Responsibilities of the position included, but were not limited by the following. As a member of the whole genome sequencing (WGS) library preparation team, our role was to ensure high quality libraries were built from the samples sent by customers from several scientific institutions. The workflow included sonication to break up the DNA. This was then visualised on an agarose gel to determine if the DNA had been broken down to the correct size. As a member of the whole genome sequencing (WGS) library preparation team, our role was to ensure high quality libraries were built from the samples sent by customers from several scientific institutions. The workflow included sonication to break up the DNA. This was then visualised on an agarose gel to determine if the DNA had been broken down to the correct size. The samples would then purified using magnetic beads before end repair. After end repair adaptors were ligated to the ends of the DNA fragments. A further purification step would occur before size selection to endure only the correctly sized DNA fragments were taken forward. Following size selection the DNA samples would be amplified by end point PCR. After amplification they samples would go through a final purification before the amount of DNA was quantified using a Qubit fluorometer.
