Research Technician Iv, Clinical Research DivisionFred Hutchinson Cancer Research Center, Jim Olson Lab, Molecular Design And Therapeutics Core Mar 2015 - Jul 2016Seattle, WaLarge scale protein and knotted peptide production and characterization using lentiviral vectors, mammalian shake flask cultures, FPLC, and SDS-PAGE. Led HPLC platform development for analysis of protein production, purification and in vivo testing groups in a fast paced, growing laboratory group. Managed two HPLC's, one paired with a Mass Spectrometer, the other in a radioactive lab utilizing a scintillation detector. Responsible for all organic and radioactive material management and disposal.

Research Scientist/Engineer Ii, Department Of NeurologyUniversity Of Washington, Jeff Chamberlain Lab, Gene Therapy Vector Core Feb 2004 - Jul 2014Seattle, WaProduced recombinant Adeno-associated virus (AAV) and Lentivirus vectors for the delivery of therapeutic or marker genes to the musculature of murine, canine, porcine, and primate animal models in the pursuit of a cure for neuromuscular disorders with an emphasis on Duchene's Muscular Dystrophy (DMD). This included all stages of the vector production from large scale plasmid preparation and characterization, cell culture and transfection (both flat culture and roller bottle), vector harvest and purification (affinity chromatography, PEG precipitation, sedimentation and isopycnic ultracentrifugation), followed by vector quantitation (reporter gene assay titers, genomic southerns, qPCR and SDS-PAGE). Responsible for the establishment and maintenance of cryo-preserved cell stocks. Provided HPLC support for members of the lab needing size exclusion chromatography and antibody production purification. Managed a dishwashing and autoclave facility for the building floor. Served as the laboratory radiation safety officer and fixed broken or installed new lab equipment. Fully responsible for the setup and management of the Vector Production Lab, including all consumables, reagents, equipment, and product distribution to collaborators. Laboratory Radiation Safety Officer.

Research Associate Iii, Cell BiologyPrimal Incorporated Dec 2000 - May 2003Seattle, WaAs the second bench scientist hired when this company was established, I was a key participant in the setup of all the wet labs and tissue culture spaces while establishing the active product pipline. Designed, implemented, and managed the use of two Tecan Gen-Mates, one Tecan Genesis robotic workstation and a Tecan PW384. These instruments were utilized in a clean room environment for both Tissue Culture and Molecular Biology methods. The primary tissue culture methods supported the generation of mutant murine Embryonic Stem (ES) Cell libraries and the associated High Throughput Screening methods for the isolation of ES clones of interest. The molecular biology methods included genomic DNA preparation and associated High Throughput nested PCR screening in both 96 and 384 well formats. Responsible for managing all DNA sequencing efforts for the Cell Biology, Molecular Biology and New Technology groups as well as DNA analysis for Cell Biology using a Applied Biosystems 3100 Genetic Analyzer and Sequencer software. Maintained and purified large scale murine ES cell genomic DNA stocks for High Throughput Screening. Cryopreservation of knockout ES Cell libraries.

Research Technologist Ii, Virology CoreUniversity Of Washington National Primate Research Center Oct 1995 - Dec 2000Seattle, WaProvided molecular biology, immunology, and cell culture support for studies focused on AIDS vaccine development in various non-human primate model systems for both academic and industrial laboratory investigators. Most work was performed under BSL-3 conditions. Responsible for the development and maintenance of in-house ELISAs. This included the growth, purification, and quantitation of monoclonal antibodies from both Tissue Culture and Ascities. During my time in this lab we relocated from the UW main campus to an Alexandria property on Western Avenue, I was responsible for setting up all the Virology Core wet lab bench spaces, BSL 2 and 3 tissue culture labs, and moving, maintaining and monitoring all the cryopreservation equipment.
