We are focused on some of the most devastating neurological and neuromuscular diseases that affect children. Together with non-profits like Hannah's Hope and FARA we are dedicated to finding therapeutic approaches for a range of diseases such as, Duchenne Muscular Dystrophy (DMD) to lesser known diseases, such as giant axonal neuropathy (GAN), Friedreich ataxia (FA), and Canavan disease.Our therapeutic approach targets the underlying genetic defects and the hope is that this will alter the course of these diseases. Through the use of highly specific recombinant AAV we are seeking to develop the processes necessary to produce effective, high-quality, vector on a large manufacturing scale.

Experience in production and purification of research grade, GLP, and cGMP grade rAAV using wave bioreactors. Maintined HEK 293 suspension cell line culture for both large and small-scale transfection. Chromatography purification of multpile serotypes of AAV. Ability to pack columns using AKTA FPLC systems and use Unicorn software for method editing or manual setup. Performed traditional restriction digest cloning and Gibson Assembly to create custom AAVplasmid expression cassettes. Experience with using VectorNTI software for primer design, cloning strageties, and sequence verification.

Independent ResearchPerformed the analysis of a RAD-seq experiment on the Shortfin mako population variation off the coast of North Carolina. Used the Stacks software pipeline for interpreting RAD-seq results and the population genetics program STRUCTURE to carry out assignment tests.Gained experience using PCR, DNA purification, and mitochondrial genome sequencing by genetically testing Scalloped hammerheads (Sphryna lewini) in the hopes of recognizing the newly identified Carolina Hammerhead (Sphyrna gilberti).CAPSTONE ResearchTeamed up with other UNC students to study the shading affects of floating docks on the ecosystem. The results were displayed in a public presentation to the owner of the private marina and community.

Ecuador

UNC GAIAS study abroad program Ecuador and Galapagos Islands Quito, Ecuador Independent Research San Cristobal, GalapagosAided in tracking and tagging juvenile Blacktip sharks (Carcharinus limbatus). Learned to safely attach acoustic transmitters and operate a hydrophone. Worked with another UNC student to create a photo database of sea turtles to be used as a source of indentification in future research. Methods involved taking high resolution photos of facial scales and the carapace to create a unique ID for each turtle. Performed a necropsy on a juvenile sea lion to help determine cause of death and aid in the investigation of a pathogen affecting the local sea lion population.
