-Optimized and conducted a series of high throughput, thermotolerance assays to screen 5000+ enzyme variants.-SDS-Page and Bradford assays. -Kinetic and endpoint enzymatic reactions. -Glucose Oxidase fluorescence assay. -Purification using hollow fiber diafiltration.-Fungal expression of proteins.

-Composed multi-fragment assemblies using type IIS restriction enzymes and three part ligation optimization.-Generated 800 DNA chimeras per week.-Programmed and utilized automation instruments including Tecan, Qpix, and Saffire. -Qiagen based plasmid DNA isolation from bacterial colonies, restriction digestion, dephosphorylation, ligation and transformation for vector construction. -PCR optimization.-Applied Sanger Sequencing method (primer extension, G50 filtration, Sequencer) and analyzed results using bioinformatic specific toolset.

-Seaward sample collection using Van Veen, CREEB coring, dredging and Niskin bottle techniques. -DNA extraction, PCR, Cycle Sequencing and Direct Sequencing in efforts to test for Paralytic Shellfish Poisoning.-Analyzed sequences using Sequencher DNA analysis program and NCBI.

-Developed, standardized, and interpreted QPCR results using ABI Prism 7000. -Performed DNA plasmid digestions and ligations with vectors including molecular cloning.-Performed Western and Southern Blots. -Used Mini and Maxi Preps for PCR reactions.-Gave several progress presentations to the department and company. -Wrote and amended several SOP’s for techniques determined by my research.
