The goal of the project was to develop a reporter mouse model to follow Adipose Stem Cells migration. I worked on the design of specific promoters, molecular cloning and transfection optimization of ASC (Nucleofection vs Lipofection) and gene expression characterization by RTqPCR, confocal microscopy and flow cytometry.

Mhlanga team “Gene expression and biophisics”:http://mhlangalab.synbio.csir.co.za/- Development of nanoprobes able to detect specific DNA sequences at low concentration by super resolution microscopy. Production and characterisation of gold nanorods, conjugation with molecular beacons, detection at the single molecule level and image processing (project in collaboration with Pr. Malik Maaza at iThemba Labs, Cape Town, South Africa).- Detection by immunofluorescence and super resolution microscopy (STORM) of HIV env proteins from the surface of infected cells.- Production of TALE nucleases constructs able to modify a specific gene.- Reprogrammation of differenciated cells into induced pluripotent cells (iPS) and redifferenciation.- Involved in organisation and teaching of super resolution microscopy courses during the EMBO Pratical Courses of 2011 and 2012 in South Africa.

Borg team “Cellular polarity, signalisation and cancer”:http://crcm.marseille.inserm.fr/equipesde-recherche/jean-paul-borg/- Monoclonal antibodies productions against human et mouse PTK7 (receptor tyrosine kinase involved in cancer process), characterisation by flow cytométrie, Western Blot, Immunoprecipitation, purification by FPLC chromatography, ascites production, antibodies isotyping and fluorochrome conjugation (cellular culture, cellular lysates, proteins quantification by Bradford, gel SDS-PAGE coloration by Coomassie blue).- Production of a soluble PTK7 receptor: cDNA obtention by RNA extraction and RT-PCR, cloning process in molecular biology.- Knowledge of IHC, oncology, embryogenesis, RTK and planar polarity.
