Custom synthesis of compound libraries and biologically active molecules. Multi-gram synthesis of catalog compounds.

Designed and produced small focused compound libraries using solution phase parallel synthesis for rapid lead optimization of several CNS programs. Synthesized library scaffolds in gram quantities through multi-step synthetic sequences. Extensive use of laboratory automation for library production. Optimized a multi-step synthetic route to generate new library scaffolds in gram quantities. Developed new methods to solve a critical synthetic step towards the formation of a thio ether bond. Synthesized several compound libraries from which key potent and selective antagonists were discovered. Developed a new synthetic route for the decarboxylation step in the synthesis of a scaffold. Established protocols for the synthesis and distribution of compound libraries on liquid handling robots.

Synthesized small focused libraries and single compounds to investigate the SAR around lead compounds in several cardiovascular and metabolic programs. Solution phase and solid phase compound libraries were produced around leads from high throughput screening. Resynthesized a lead from high throughput screening in an early phase cardiovascular program to confirm its structure and activity.Synthesized a new lead compound in an early phase cardiovascular program. Optimized a critical Mitsunobu coupling in a metabolic program. Produced a 100 member combinatorial library of a lead benzimidazole chemotype.Resynthesized two active compounds in a late phase metabolic program for profiling and in vivo studies.Synthesized a small multi-step solution phase library to investigate amidine replacements in a lead compound in a cardiovascular program.Prepared a compound library for an early phase cardiovascular program which provided SAR and selectivity data for the lead chemotype.

Produced several large compound libraries for general screening in house. Developed new reactions on solid phase supports for library synthesis. Designed protocols for solid phase library production using Irori technology. Produced small lead optimization libraries for a cardiovascular program by both parallel solution phase and solid phase synthesis. Produced both a 15K member benzothiazepine library and a 10K member piperazine-2-carboxamide library using Irori technology. Developed a new isonitrile resin to produce carboxylic acids from the Ugi reaction.

Synthesized lead compounds in multi-gram quantities for animal testing. Resynthesized key intermediates in large scale for use by other medicinal chemists. Synthesized reference competitor compounds.

Summer Intern – Quality Control (1986, 1987 and1989) Performed assays for the quantitative determination of active ingredient in pharmaceutical products. Ran UV-VIS, KF, Heavy Metal, Color, pH and other assays on pharmaceutical products.
