For dry powder inhaler (DPI) formulations, the interactive forces between the API and excipient impact on the performance of the formulation. Furthermore, the vast mechanical stresses subjected to the API during mechanical processing, such as micronization, combined with the relative inefficiency of the this operation highlight the importance of understanding the properties of the primary crystalline feed material in order to produce materials for inclusion in the final drug product with defined specification and functionality. The Young’s modulus represents a material’s resistance to elastic deformation and is known to have a strong influence on the micronization behaviour of solids while the use of different solvents and anti-solvents are known to affect the kinetics of crystal growth and subsequently modify the crystal habit. This work aimed to evaluate the relationship between the primary crystallization of the API and the impact modifications of this process have on the secondary processing of these materials. Specifically, the project addressed the mechanical properties of the crystals produced and the influence these properties had on the milling and performance of DPI products.
