The title of this internship is called "Enzymatic and Kinetic Studies of Protein-Ligand Interactions". Expressed and purified a heme-containing enzyme which is an important target in cancer and neurodegenerative diseases. The goal was to develop an assay to study the interaction of inhibitors with the enzyme in different states. Once the enzyme was purified using ÄKTA purification system, the enzyme was used to study enzyme functionality/ inhibition with discontinuous and continuous enzyme assays. Product formation by the enzyme was analyzed. Interaction analysis between the enzyme and ligands was performed using Surface Plasmon Resonance biosensor technology (SPR).I gained experimental and theoretical knowledge in the following techniques: E. coli Protein Expression & Purification, Steady state Enzymatic Assays and Surface Plasmon Resonance Biosensor Technology.
