PCR: Developed two-step PCR conditions for optimal Genotyping analysis. Optimized PCR cleanup for Mass spectrometer analysis. Routinely and reproducibly performed low volume PCR reactions during development experimentation as well as on client samples in the 384 well format. Operated Cartesian Technologies Microarrayer for small volume PCR reactions.CLONING: Performed digestion and ligation on a daily basis. Optimized ligation protocol for technology specific applications. Separated fragments via agarose gel, PAGE, or HPLC purification. Transformed electrocompetent cells with plasmid DNA. Picked and grew colonies for later plasmid preparation. Performed plasmid preps and user maintenance on the Qiarobot 8000. MASS SPECTROMETER: Evaluated and implemented Chemstation software with the CTC well plate autosampler. Created and integrated the Mass spectrometer Calibration and Quality Control protocols. Maintained the Mass spectrometer at the user and engineer levels while providing in house technical support.HPLC:Maintained HPLC at the operator level. Set up and prepared samples for automated sample collection. Performed fragment analysis to determine the correct sample for processing.SEQUENCING: Set up and amplified sequencing reactions at 1/16th volume. Separated amplicon from Big Dye nucleotides using magnetic bead hybridization. Operated and performed user maintenance on the ABI sequencers.PHARMACOGENOMICS & WOMEN’S HEALTH: Using Roche MagnaPure 96 and Promega Maxwell 16 systems, extracted client DNA from biological samples including: buccal swabs, thin preps, and blood. Using Genmark, Autogenomics, and Luminex platforms, set up amplification reactions as well as post PCR processing. Programmed and operated Beckman Multimek automated pipetting system. Operated Roche Cobas x480 automated sample processing system.
