Dedicated research work to explore the potential for small molecules, called foldamers, to mimic portions of proteins’ 3D shape and behaviour, towards generating hybrid “bionic” proteins, with a long-term aim of developing new therapeutics.Full abstract:De novo design of foldamers is a current challenge of chemical biology, and overcoming it is essential to expand the protein toolbox. α-helix mimicry is a key approach to building foldamers and a stepping-stone towards generating "bionic proteins", i.e. proteins comprising non-natural segments, with enhanced biological functions. As part of this aim, my work as a PhD student in the Wilson Group focused on using proteomimetics to build novel 3-dimensional constructs.As a groundwork towards generating proteomimetic-based coiled coils, I designed and synthesised small aromatic oligoamides and carried out extensive NMR studies to assess their ability to form supramolecular complexes through self-assembly. I also investigated the interactions between aromatic oligoamides and Affimers, a non-antibody-based scaffold developped in the BioScreening and Technology Group at the University of Leeds. In tandem with phage display, I screened a library of Affimers against a small library of biotinylated N-alkylated proteomimetics and characterised several types of recognitions with ELISA and Fluorescence Anisotropy.I presented my results at internal and international conferences and published 2 first-author papers.
