Initiated project aimed at streamlining process for identifying potentially viable male contraceptive drugs using published mouse models, DNA-Encoded Chemistry Drug Libraries, Solubility Assays, and Computer Automated Sperm Analysis and Sperm Motility Assays.Responsibilities included literature research, protocol design and optimization, data manipulation and statistical analysis, observing experiments virtually, and participation in daily research seminars, weekly journal clubs and drug selection meetings, and a series of practical lab skill lectures.Successfully optimized a universal drug solubility assay and sperm-analysis assay to allow for efficient evaluation of solubility and efficacy of drugs identified within DEC Libraries for the Center for Drug Discovery.

Investigated behavior and characteristics of specific bacterial enzyme, Topoisomerase IV, and the manner and speed at which it catenated DNA in varying topological states.Applied enzymatic time course assays, assay optimization and reactant titrations, SDS-PAGE gel analysis, DNA quantification, statistical analysis, and essential lab skills.Studied and discussed literature covering DNA topology, coildness, catenation, de-tangling and unwinding, specific enzyme structure and function, and chemotherapeutic and antibacterial drugs and poisons involved in topoisomerase inhibition.Determined that Topoisomerase IV has a supercoil handedness preference for catenating positively coiled DNA above negatively coiled or relaxed DNA.

Designed and executed full research project in collaboration with another intern focused on the effect of melatonin production on a house sparrow’s gut microbiome through tissue sampling and lysis, microbial DNA collection, DNA purification and specking, and Quantitative PCR.Participated in weekly lab meetings, program seminars, and mini courses on lab basics including lab notebook upkeep, dilutions, important calculations, and other essential lab skills.Revealed that microbial circadian rhythms may be independent of the organism’s overall circadian rhythm through quantification of microbial DNA present in samples from pinealectomized (disrupted circadian rhythm) and sham birds at varying timepoints and meals.
