The aim of this study was to determine the kinetics of TNF-like ligand1A (TL1A) mRNA expression and protein production in monocytes and monocyte-derived dendritic cells and to assess whether certain interleukins have the ability to control TL1A in these antigen presenting cells. To determine soluble TL1A protein, an enzyme-linked immunosorbent assay was set up. Membrane bound TL1A was detected by a newly developed flowcytometric analysis and TL1A mRNA was detected by quantitative SYBR green PCR.Techniques: Mammalian cell culture, ELISA, immunohistochemistry, flow cytometry, qPCR.

This research project focused on quantitative steady-state pH measurements in the apicoplast of Plasmodium falciparum blood stage parasites. The apicoplast is a characteristic organelle of Plasmodium spp. and its function is fundamental to the parasite’s survival. The unusual origin of the relict plastid with its non-mammalian metabolism and transport pathways presents attractive new strategies for prospective drug targets. We generated a construct that transiently expresses the bipartite signal sequence of the acyl carrier protein fused to the pH-sensitive green fluorescent protein pHluorin. We successfully localized the recombinant protein to this organelle and performed quantitative pH determinations.Techniques: DNA cloning, culture and transfection of P. falciparum, confocal live-cell fluorescence microscopy.
