Development Of Thiourea Derivatives As Antiviral Agent For Hepatitis B Virus Infection
Interference With Hepatitis B Virus Replication And Viral Gene Expression By Thiourea Derivatives
Graduate Student, The Institute of Liver and Biliary Sciences, New Delhi, India.Dept. of Molecular & Cellular Medicine, (Prof. Vijay Kumar),Ph.D. thesis “Interference with Hepatitis B virus replication and viral gene and expression by thioureaderivatives”.Research Experience: We have initiated a project to find new ways to treat Chronic Hepatitis B infection. So far, we have developed and patented three thiourea derivatives out of twenty, that effective against HBV as antivirals. We also reported the interaction of these thiourea derivatives with target proteins (HBx) of HBV by Molecular Docking. The antiviral properties of these derivatives are comparable to standard drugs like tenofovir and entecavir, and along with these, they show additive effects. Therefore, because these derivatives have a distinct mode of action from currently available drugs, they are suitable for combination therapy. Further, we evaluated the pharmacokinetics, pharmacodynamics, and oral and intravenous bioavailability of effective derivatives in a rodent model (Rat) by Mass spectrometry.Additionally, I independently performed a pilot study on "the effect of thiourea derivatives onchronically infected hepatocytes" and found that, in contrast to Entecavir and Vitamin C, thiourea derivatives treated hepatocytes increased the expression of antiviral proteins and oxidative phosphorylation. Entecavir and vitamin C are used as positive controls because of their antiviral and antioxidant properties respectively.Supervisory Experience: Supervised two masters students, undergraduate student, and three graduate students. Took care of Molecular biology cell culture lab and proteomics facilities.

Research Experience: With Dr. Anupam Kumar, I was involved with a project on the Bone marrow response after G-CSF and Mesenchymal Stem cell therapy in Chronic liver disease patients and found that after therapy, patients recovered quickly.With Dr. JS Maras, involved with two projects on the role of Iron (Fe++) accumulation in thehepatocytes and macrophages of Severe alcoholic hepatitis (SAH) patients, we found iron mediatesinflammation through ADAM17 induction, resulting in macrophage activation and increased sheddingof TNF-α and sCD163. In another project, changes in urine albumin were used as a molecular marker tofind SAH. We found that changes in urine albumin reflect changes in blood albumin, and that averageurine albumin levels could be used to predict how steroids therapy works and the mortality of SAH patients.

Khan & Dr. Anand Prakash)Research Experience: With Dr. Anand Prakash, I have completed my master’s dissertation on theproject “the role of cyclic AMP Response Element Binding (CREB) proteins in Prefrontal Cortex andAmygdala during Fear Memory Consolidation and Extinction”. This study looks at the role of CREB inthese parts of the brain. CREB was linked to neuronal activation and was much lower in the infralimbicprefrontal cortex of the rapid extinction group than in the delayed extinction group. In conclusion, thedeficits seen in the rapid extinction group may be caused by changes in histone acetylation, which mayin turn be caused by changes in the activation of infralimbic prefrontal cortex.
