Senior Principal Scientist
CurrentSupporting covalent drug discovery efforts and the rapid exploration of new modalities by identifying ligandable pockets and chemical starting points for challenging targets using cysteine reactivity profiling, and direct-target enrichment via alkynylated probes. Developing targeted assays (MS and non-MS based) for absolute quantitation of exploratory targets and cysteine site occupancies for high-throughput in-cell engagement profiling. Employ data-independent acquisition to drive profiling of electrophilic-alkyne libraries to inform library buildout and prioritize probes for follow-up studies. Explored enzyme mediated proximity labeling as a tool for target identification using chimeric bi-functional chemical probes.
Investigator Iii
Proteomics lead for both small and large molecule target ID efforts using photo-affinity labeling, activity-based protein profiling (ABPP), chemical/photo-crosslinking, and kinase selectivity profiling. Member of target ID mechanism of action (TIMoA) team focused on identification of novel targets from phenotypic cellular screens. Employed peroxidase and biotin-ligase based proximity labeling for receptor deorphinization efforts and protein interaction mapping. Additionally, supported targeted protein degradation efforts with validation and selectivity profiling.
Investigator I & Ii
Supported small and large molecule drug discovery through mass spectrometry (MS) based protein characterization including intact mass, post-translational modification analyses, and tandem MS based peptide identification/quantification strategies. Additionally, employed structural MS for conformational dynamics, interaction mapping, and stability studies using hydrogen deuterium exchange MS (HDxMS) and chemical footprinting where we mapped protein-protein and protein-ligand interactions including antibody (epitope mapping), peptide, small molecule, and aptamers for numerous programs. Led site-wide intact protein LC-MS and structural MS services.
Postdoctoral Research Assistant
Developed novel data acquisition strategy (data-independent acquisition, DIA) for improved quantification of isotopically labeled and label free samples. Designed and developed strategies for the identification and quantification of complex protein mixtures using liquid chromatography and tandem mass spectrometry. Focused on metabolic stable isotope labeling strategies (15N, SILAC, 18O) for quantification of proteins in tissue cultures, model organisms, and mice. Assisted with collaborations to identify components of complex protein mixtures using multidimensional liquid chromatography coupled to tandem mass spectrometry (MudPIT).
Research Assistant
Focused on fundamental studies in quantitative mass spectrometry using inductively coupled plasma mass spectrometry (ICPMS). Investigated analyte transport to the mass spectrometer from sample introduction sources. Designed and built a novel multiplexed electrothermal vaporization (ETV) system for high throughput sample introduction. Developed method for optimization of multi-element quantification using a scanning ICPMS.
Teaching Assistant
Instructed upper division quantitative analysis labs and introductory chemistry discussion sections for chemistry majors and non-majors.
Research Assistant
Investigated DNA regulation and characterization among ciliated protozoa.
Research Assistant
Studied receptor binding and adenosine transport within vitamin B6 deficient mice
