Student InternScelse Jan 2015 - May 2015SingaporeI was involved in two mini projects during my student intern period and had acquired molecular techniques such as primers construction, polymerase chain reaction (PCR), gel electrophoresis, minimal inhibition concentration (MIC) assay, statistical and data analysis,as well as data presentation. Below are the summary of the mini-projects : (I) Identification of efflux pump inhibitor (EPI) that were synergistic to antibioticEfflux system is one the protecting mechanism in bacteria, its function is to extrude the harmful substances that serve to threaten survival of the bacteria. The control of efflux system is also the new directions to restore the activity of antibiotic. EPI was previously reported to restore the ineffective ciprofloxacin function on pseudomonas aeruginosa and was found to be effective in suppressing MDR. The aim of this study is to identify for effective and potential efflux pump inhibitors (EPI) that can be use synergically with antibiotic, to ultimately lower the minimal inhibition concentration (MIC) of antibiotic in treatment to inhibit bacterial infection. (II) Construction of knockout mutant in pseudomonas aeruginosa The construction of knockout mutant enables the discovery of the function of gene of interest in the host body and this provide mutant strains for downstream experimental assay. The traditional method of gene replacement can be tedious and laborious; hence, the multisite gateway method was performed to provide shorter time and greater efficiency at constructing a knockout through transferring heterologous DNA into multiple vector system.
