Site leader in microscopy: devised and implemented fluorescent imaging and analysis techniques specific to the development of Ion Torrent sequencing platforms, including characterization of DNA-templated beads and quantifying the success of loading beads into microwells on a surface. Maintained a set of current protocols for all existing techniques and kept abreast of needs for all projects at the site, as well as receiving samples from other locations for specialized analyses. Sensor development team member: tested different semiconductor sensor configurations and investigated interactions among the sensor build, chemical surface treatment, and DNA bead composition to select optimal designs. Testing was done with full sequencing runs using the Ion Torrent instrument suite in customer configurations, as well as using internal quality control configurations and instrumentation. Projects included development of new products and testing instruments, and refinement of released products.

Groton, Ct

Conducted field campaigns to collect oceanographic data in Florida Bay and in Elkhorn Slough, California. Used radiometers to measure properties of light above and below the water surface, and used in-situ particle analyzers to characterize the water column. Coordinated work in Elkhorn Slough to coincide with overflights of NASA’s PRISM imager to deliver a complete package of hyperspectral aerial image data with ground-truth verification data. Laboratory analysis of seagrass samples to calculate Leaf Area Index metric for the sampling sites, and of water samples to calculate chlorophyll content in the water column at different locations of interest. Synthesized the ground-truth verification data from several radiometers and particle analyzers and used this data to model the water column and calculate the expected remote sensing reflectance for different benthic habitat types. These models as well as image classification techniques were used to analyze the aerial imagery and determine the benthic habitat for each pixel in the image data.
