Project lead on programs aimed at drugging E3 ubiquitin ligases, including the first prospective discovery of molecular glue small molecules that enhance the interaction between an E3 ligase and its substrate. We employed high-throughput screening and structure-based drug design to identify and optimize small molecules that bind at the protein-protein interface of the E3 ligase ß-TrCP and its substrate ß-Catenin, improving the binding affinity of degradation resistant ß-Catenin for ß-TrCP over 1500-fold, causing its ubiquitylation and degradation. This approach of restoring degradative function by identifying molecular glue that enhances protein-protein interactions disrupted in disease is complementary to the bifunctional PROTAC approach of induced protein degradation and can be applied to many other difficult-to-drug disease targets.Project leadership and support for various lead discovery programs through: • Target identification and validation• Protein design and purification• Fluorescence-based protein-protein interaction assay development• Biophysical assay development• Enzymatic (ubiquitylation) assay development• High-throughput screening• X-ray crystallography (crystal growth, data collection, structure solution, model building)• Cellular target engagement assay development• Close collaboration and communication with medicinal and computational chemistry
