Postdoctoral Research Scholar (Hurley Lab)

Adaptor protein structure and function in trafficking disregulated disease states, such as in HIV infection (Hurley lab).I continued to grow my skills in structural biology by cryo-EM, now working on determining the high resolution structure of the trimeric macro molecular complex of Arf1:AP-1:HIV-Nef:Tetherin by cryo-EM. This work was published in Morris et al Cell 2018.My scripting and Linux environment development proved highly effective resulting in the construction and set up of 4 GPU servers providing all image processing capacity for the lab. I continued to code to tackle unique image processing problems but in this role aimed at creating more generalised programs and workflows that others can use in their research. The is reflected in my GitHub account www.github.com/kylelmorris. I also worked highly collaboratively and supportively of colleagues around me, leading to the successful publication of three additional EM structures of proteins involved in autophagy, nutrient and soluble gas sensing. Training and mentoring further brought at least two new researchers successfully into the field.

Postdoctoral Research Fellow (Smith Lab)

Clathrin assembly and disassembly, structure and regulation of self-assembly (Smith lab). I developed a structure determination cryo-EM project that has lead to the highest resolution structure of a clathrin cage to date. This analysis also revealed a number of cage architectures, some of which are newly discovered. Taken together we disseminated new insight into the self-assembly of this fascinating system (Morris et al NSMB 2019).Day to day work included classic endogenous purification biochemistry, recombinant work, all EM grid optimisation at room temperature and cryo conditions, and lots of travel around the world to the early Titan Krios/direct electron detector microscopes that existed at the time. As part of this work I continued to develop my programming skills, now scripting heavily in Linux shell (bash) and to a lesser extent python to develop image processing workflows.I also developed single molecule FRET based assays (confocal ALEX FRET) to monitor clathrin cage assembly and disassembly, as well as adaptor binding and mediated cage disassembly.
