The research focused on developing in vivo approaches, including transient GFP markers and siRNA knockdowns, for studying neurokinin B (NKB) and the NKB receptor (NK3) in the rat brain. Studies included the intra-cellular trafficking and compartmentalization of NK3 within the Paraventricular Nucleus (PVN). Specifically, designed, purified, and characterized new polyclonal antibodies against both NK3 and NKB. Transcriptional studies required the development of micro-punch real time PCR procedures to measure NK3 and NKB transcripts. Initiated immuno-coprecipitation studies to begin to characterize chaperone proteins involved with NK3 function in vivo. siRNA knockdown studies were started to disrupt the NK3 receptor system in vivo to identify NK3’s role in fluid balance within the PVN.
