Master's thesis - "Evolution of an iron-detoxifying protein: uncovering the function of a group of conserved amino acids in eukaryotic frataxins"During my master's practical work, I gained hands-on experience in growing bacterial and yeast cultures and in protein engineering, including transformation, expression and purification. I used electrophoretic techniques such as SDS-PAGE and native electrophoresis, along with Coomassie Brilliant Blue staining and Western Blotting, to analyse proteins. I assessed the stability of the proteins using thermal shift assays and intrinsic fluorescence emission studies. I also assessed ferroxidase activity using iron quantification, kinetic analysis by spectrophotometer and the ferrozine assay. These experiences provided me with essential skills for biological and biochemical research.

Preparation and analysis of various samples using instrumental techniques such as HPLC-DAD, CG-FID, UV-Vis spectrophotometry, manual and automatic titrations. Preparation of spreadsheets for control and calculation of analyzed results. Support for the development and validation of analytical methods.

During my time with the Oxidative Stress Biochemistry Research Group, I was involved in analyzing the expression levels of various proteins related to energy metabolism using diverse biological models, ranging from yeast to human samples. The primary analytical approach utilized in these studies was targeted proteomics based on SRM (Selected Reaction Monitoring).Throughout this work, I gained valuable practical experience in tasks such as preparing cell lysates, quantifying total protein content, digesting protein samples, and employing triple quadrupole mass spectrometry coupled with liquid chromatography for sample analysis. Additionally, I actively engaged in activities related to yeast cultures and other essential laboratory techniques such as western blotting and cell culture.

Activities for preparing and monitoring classes in the chemistry cycle (inorganic, organic, analytical, electrochemical and corrosion), chemical engineering (characterization of materials, unit operations) and food engineering (biochemistry, biology, microbiology, sensory analysis). In addition to the quality management of the sector, application of 5S, people management and management of chemical waste generated in the laboratory.

Performance in the Governance group (Smart Campus Facens - SCF), which manages and provides support to the other groups (Health and Quality of Life; Energy; Environment; ICT; Education and Culture; Mobility and Security; Urbanization; Industry and Business) . In addition, tasks related to the management of communication, of the projects developed by the SCF, from social media, electronic media and academic events were assigned.
