In my project I examined how vitrification affects the chromosomal status of murine embryos. According to my project, the embryos were vitrified at morula/blastocyst stage, inserted into culture to recover and the following day the chromosomal constitution of the blastocysts was analysed. The method which was used for this was Fluorescent In Situ Hybridization (FISH). The results were compared to those of the control group which consisted of non-vitrified embryos in order to understand whether vitrification was the factor that caused these chromosomal abnormalities to the blastocysts. Total genomic DNA was isolated from the organism. The desired area was amplified through PCR by using the appropriate primers, followed by sequencing of the desirable fragment. After the sequencing results were edited and were submitted to Barcoding of Life Database (BOLD). The obtained results were very interesting and led to publication of a scientific paper in 2011 in a special edition for sequencing of the mitochondrial DNA. Techniques learned: DNA extraction, PCR, sequencing, DNA electrophoresis, data analysis, submission of the results to the BOLD database.This whole year much research and lab experience was gained.Dissertation’s final degree: 10/10
