The research project performed during and after my masters thesis studies aimed at characterizing the thermodynamics of interaction between the cytokine hormone prolactin and its cognate receptor. A number of eleven prolactin mutants (receptor agonist and antagonists) were designed in order to alter the pH dependence of the receptor binding, as well affinity optimize the interaction at physiolocical pH as well as acidic endosomal pH. The prolactin mutants, and extracellular part of the prolactin receptor were cloned, expressed, and purified from inclusion bodies (bulk and chromatography techniques) and characterized using a combination of circular dichroism spectroscopy, fluorescence spectroscopy, MALDI TOF MS, Native PAGE, and Isoelectric focusing. The thermodynamics of the prolactin mutants interaction with the extracellular part of the prolactin receptor were characterized using isothermal titration calorimetry and surface plasmon resonance.
